Overview
This service provides whole-genome sequencing using the Sequel IIe or Revio system. Users submit genomic DNA or metagenomic samples, and SignAC prepares libraries using the PacBio SMRTbell Prep Kit 3.0 from the samples and generates HiFi reads using the Sequel IIe or Revio system.

Sample submission guidelines
Genomic DNA
- Volume: minimum 8 µl
- Total amount: minimum 7 µg
- Concentration: minimum 60 ng/µl
※Please quantify your sample using a Qubit fluorometer.
※We recommend high-molecular-weight DNA, that is, >50 kb (70% ≧ 10 kb; GQN10kb ≧ 7.0 and 50% ≧ 30 kb; GQN30kb ≧ 5.0)
※We recommend the quality check using Femto Pulse. SignAC offers QC service (link).
※The presence of contaminants may adversely affect library preparation.
※If the genomic DNA sample has a high level of DNA damage, sufficient data may not be obtained.
※Please submit your samples in 1.5 mL tubes. We recommend using DNA low-bind tubes.
Data delivery

- Output: ~30 Gb (Sequel IIe)/~90 Gb (Revio)
We do not guarantee the data amount. - Please note that this service provides analysis on a per-SMRT cell basis. We offer “library preparation only” as an additional option, allowing for multiplex analysis of multiple libraries within a single SMRT cell.
