Overview
This service provides full-length RNA sequence analysis using the Sequel IIe or Revio system. Users submit total RNA samples, and SignAC prepares libraries using the PacBio Iso-seq library and generates HiFi reads.

Sample submission guidelines
Total RNA
- Volume: minimum 8 µl
- Total amount: minimum 700 ng
- Concentration: minimum 60 ng/µl
- Please quantify your sample using a Qubit fluorometer.
- ≥7.0 RIN
- Because reverse transcription is performed using oligo(dT) primers, RNA from organisms lacking poly(A) tails cannot be analyzed.
- Please submit your samples in 1.5 ml tubes. We recommend using DNA low-bind tubes.
Data delivery
hifi_reads.bam files
- Output: ~2 million reads (Sequel IIe)/~6 million reads (Revio)
We do not guarantee the data amount. - Please note that this service provides analysis on a per-SMRT cell basis. We offer “library preparation only” as an additional option, allowing for multiplex analysis of multiple libraries within a single SMRT cell.
