Overview

This service provides an ONT rapid sequencing kit (SQK-RAD114) and DNA sequencing analysis using PromethION24 (Oxford Nanopore Technologies). Customers are required to submit DNA samples (genomic DNA, amplicon, etc.), and we prepare a library, load it onto an R10.4.1 flow cell once, perform sequencing, and deliver the sequencing data.

The omission of purification steps allows for lower input DNA amounts.

Sample submission guidelines

DNA sample (>300 ng)

Please contact us if you can only provide samples of smaller amounts.
We recommend gDNA with OD_260/280 > 1.8 and OD_260/230 > 2.0–2.2 as measured by Nanodrop.

※Please submit your samples in 1.5 mL tubes. We recommend using DNA low-bind tubes.

Please submit an Excel file with the sample name (information written on the tube) and concentration through the Redmine system.

Add a serial number to the sample name and write the number to the tube as well.
As a general rule, proceed with library preparation based on the concentration specified by each individual.
Please write your name and the Redmine ticket number on your sample tube.

Data delivery

POD5/fastq files

Output: 25–100 Gb/single flow cell.
We do not guarantee the data amount.
The sequencing portion is a per-flow cell analysis service. You can also add the “library preparation only” option to multiplex multiple libraries within a single flow cell.
FASTQ files will be generated by the latest version of MinKnow’s default Basecaller with high-accuracy mode.
Please contact us if you need 5mC detection data.
